Leon S. Kennedy
At first Ada was unfriendly with Leon, but her manner softened when they teamed up and got to know one another better. Ada was touched by Leon's kindness and also greatly respected his intelligence and skill. Something about Leon stirred warm feelings within her and she unexpectedly fell in love with him. Since their first meeting in Raccoon City, the two have been able to forget each other and have kept tabs on one another.
Months before the events of Resident Evil 4, Ada found out that Leon was assigned to rescue the president's daughter from a religious cult located in a small town in Spain. Being already involved, she knew that she would have to aid and guide Leon behind the scenes so that he could get out of the village safely. At the end of RE4, Leon mentions to Ashley that Ada is like a part of him he can't let go. Whether they're working together or separately, they always carry their feelings for one another with them.


Albert Wesker
Ada has known Wesker for a long time, though how and when they met is a mystery. Wesker hired Ada to retrieve the G-Virus sample from Raccoon City in 1998. He watched as Ada developed feelings for Leon and, although he believed her to have failed in her mission by doing so, saved her life because he felt there was still a use for her. Six years later he hired her again in an effort to obtain a Plaga sample from a religious cult in Spain. Ada's true affiliation is with another organization, however, so she dupes Wesker by sending him something else. Both of them seem to have this understanding that the other is not trustworthy, yet they will continue to play the game until the final resolution, whatever that may be.


Jack Krauser
Krauser has been the subject of extensive research by Ada's organization and Ada concludes that he's a great soldier, but not much more than that. Wesker chose Krauser to retrieve the sample, but he ran into some unexpected problems and had no choice but to call on Ada for assistance. Even so, he dislikes and distrusts her, so much that he makes plans to kill her along with Leon. After Ada rescues Leon from Krauser, he realizes that the two are in some way connected and would probably have used this knowledge against her had she not disposed of him.


Luis Sera
In order to gain access to the Plaga sample easily, someone who was trusted by the cult was needed. Ada intercepted an email for help Luis sent to an old colleague and decided to help him out because she liked him and could relate with his situation. He was desperate to get out of Spain and agreed to fetch a sample of Las Plagas in exchange for the removal of the Plaga egg in his body and aid in his escape. Luis finds Ada very attractive and flirts around with her, but Ada always maintains her businesslike manner, even though she truly has an affinity for him. They were supposed to meet up once Luis retrieved the sample, but Luis was killed and the sample taken from him before they could.


Sherry Birkin
Since Wesker has Sherry in his custody, this could also mean that Ada has had contact with the girl. On what level, I don't know. It could merely mean that she has seen her or it could even suggest that Ada helps to care for Sherry. Other than that, the only solid connection between the two would be that Ada has Sherry's gold locket in her possession.


Annette Birkin
It was Annette who broke the news to Ada that her boyfriend, John, had died, but not sympathetically. Though she was edgy with just about everyone in the game, Annette was definitely not pleased to see Ada, especially after she discovered she was a spy sent to intercept her husband's precious G-Virus. She was so bitter about this in fact, that she attempted murder on Ada twice. Ada didn't seem to care too much for Annette either, since she pushed her off a ramp during a struggle.


John
Ada dated this Umbrella scientist in order to get inside information on the viruses being produced within the company. John entrusted her with his username and password to access information on his work, leading me to believe that he actually did care for Ada. We never get to find out if the feelings were mutual, but I do think they were. After Ada discovers John is dead, she mentions it to Leon and then quickly puts the subject aside sort of sadly. It's most certainly a possibility that she eventually learned to care for him. After all, they did spend a fair amount of time with one another.
